o t bstract he purpose of this study was to evaluate the antibacerial activity of leachable components of selected rootnd filling materials: amalgam, ProRoot MTA (mineral rioxide aggregate), Intermediate Restorative Material IRM), Super Bond C&B, Geristore, Dyract, Clearfil APX omposite with SE Bond, or Protect Bond. The direct ontact test (DCT) with Staphylococcus aureus, Enterooccus faecalis, and Pseudomonas aeruginosa, was sed. The materials were tested immediately after aplication to the microtiter wells (fresh samples) and fter setting for 3 days (set samples). Ten microliters of acterial suspension was added to each well for direct ontact with each material for 1 h at 37°C. Growth of urviving bacteria was then measured in a microplate pectrophotometer hourly at 620 nm for 15 h. Twelve ncoated wells using identical inoculum size served as ositive controls. The data obtained at the end of 15 h as subjected to one-way ANOVA and post hoc comarisons were done using Tamhane’s T2 test. Fresh amples of all materials showed a 3-h delay in expoential growth of both E. faecalis and S. aureus, and a -h delay in growth of P. aeruginosa. Set samples of RM and ProRoot MTA cements showed generally reater antibacterial activity than the other materials: oth completely inhibited P. aeruginosa, and both deayed or limited growth of E. faecalis. The DCT, by being uantitative and virtually independent of solubility and iffusion, was found suitable to assay solid root-end illing materials. IRM and ProRoot MTA were generally ore potent inhibitors of bacterial-growth than the ther tested materials. (J Endod 2006;32:345–349)
